چکیده

Phorbol-14-myristate-13-acetate (PMA) (10-10 mol/l) inhibited the Ca-dependent K efflux (the Gárdos effect GE) induced by Ca, the hyperpolarisation accompanying the GE, the vanadate-induced Ca influx, and depolarised the membrane, in vanadate-treated human red blood cells (RBC). The GE induced by propranolol (PLL) was not inhibited by PMA. Both PMA and PLL stimulated the basal Ca influx. These results suggest that a) protein kinase C activity prevents the activation of GE by vanadate but PLL bypasses this mechanism, b) the stimulation of the Ca influx by PMA and the GE inhibition are caused by the membrane depolarisation, c) the basal Ca influx in human RBC is regulated in a complex manner, and d) the effect of vanadate resembles to the activation of agoniststimulated signalling pathway in non-excitable cells.

I-7: Maternal Signalling to the Placenta

Background: Though it is well established that maternal blood-borne signals influence highly the growth of the placenta, the mechanisms are not known. In vitro trophoblast culture models are limited by an inability to reconstruct the polarised bilayer of the human hemochorial placenta. We have used a first trimester villous tissue explant system to investigate how growth factors interact with p...

I-34: Steroid Hormone Signalling at the FetomaternalInterface

Background: Progesterone is indispensable for differentiation of human endometrial stromal cells (HESCs) into decidual cells, a process that critically controls embryo implantation. However, HESCs also abundantly express androgen receptors (AR), yet the role of this member of the superfamily of ligand-dependent transcription factors in the decidual process remains poorly elucidated. Materials a...

Apoptosis: from Signalling Pathways to Therapeutic Tools

Apoptosis or programmed cell death is a gene regulated phenomenon which is important in both physiological and pathological conditions. It is characterized by distinct morphological features including chromatin condensation, cell and nuclear shrinkage, membrane blebbing and oligonucleosomal DNA fragmentation. Although, two major apoptotic pathways including 1) the death receptor (extrinsic) and...

Identification of signalling cascades involved in red blood cell shrinkage and vesiculation

Even though red blood cell (RBC) vesiculation is a well-documented phenomenon, notably in the context of RBC aging and blood transfusion, the exact signalling pathways and kinases involved in this process remain largely unknown. We have established a screening method for RBC vesicle shedding using the Ca(2+) ionophore ionomycin which is a rapid and efficient method to promote vesiculation. In o...

MiR-490-5p Functions as an OncomiR in Breast Cancer by Targeting NFATc4

Breast cancer is a serious health problem worldwide in women. MicroRNAs are small non-coding RNAs of 18–25 nucleotides in length that post-transcriptionally modulate gene expression. MiR-490 has been reported as a tumor suppressor and oncomiR microRNA in breast cancer with two separate targets, NFAT and Rho. NFAT is one of the targets for miR-490 but the relationship between hsa</e...
